General Scientific Interest:
The orofacial sphere, and in particular the mouth, is subject to highly prevalent chronic pathologies, such as dental caries which can lead to jaw bone infection, as well as periodontal diseases (i.e. inflammation and destruction of the gum and alveolar bone). The damage caused by these diseases is a considerable burden for a large part of the population. Much remains to be done to better understand these diseases in order to propose innovative, more effective and less intrusive treatments. In this context, URP2496 is mainly interested in the mechanisms responsible for susceptibility to these diseases (defect in the formation and mineralization of the tooth and its anchoring tissues, including the jawbone), as well as in the specificities of the infection and inflammation involved in these diseases. Our goal is to use the knowledge acquired to propose concrete therapeutic solutions to patients.
Whithin the URP2496 headed by Catherine Chaussain (PUPH-DDS), the group Inflammation and chronic oral diseases (caries, periodontitis) is led by Jérôme Bouchet (MCF-HDR) and Marjolaine Gosset (PUPH-DDS). This axis is absolutely fundamental, these affections being classified by the WHO as a scourge affecting almost all populations in France, Europe and throughout the world with a strong impact on the quality of life and on comorbidities. This axis will combine the strengths of the UFR in fundamental, translational and clinical research on pulp and/or periodontal inflammation in response to infection using cellular models (pulp organoids, inflammatory cell/bone cell co-cultures), preclinical models of pulp and periodontal inflammation/infection perfectly mastered by our researchers, biological collections of gums and biological fluids (blood, saliva), cohorts of patients in connection with CRESS (pediatric cohorts (Elfe, Epipage)) for caries, and general epidemiological cohorts (Constances) or specific cohorts of chronic diseases (ComPaRe). It should be noted that these pathologies have a strong impact on general health and we are already involved in interventional clinical projects in cardiology (RHU iVasc) and in IBD (Periogut), as well as in observational studies of patients with chronic diseases (cystic fibrosis, Sjögren’s disease, Wilson’s disease and other copper diseases) with the reference centers for these diseases. Finally, the group is involved in several scientific consortia, including the FHU PaCeMM, the Labex “Inflamex”.
Material and technology:
The URP2496 laboratory houses on the Montrouge site high-resolution micro-scanners (Micro-CT) imaging equipment available, through a technical platform, to the whole scientific community. The Micro-CT platform is attached to the Life Imaging platform (PIV) labelled IBISA and attached to the Paris center node of the France Life Imaging infrastructure (FLI). We currently have on the small animal X-ray imaging platform two Micro-scanners : the Quantum FX which is an in vivo Micro-CT commercialized by the company PerkinElmer® and the Skyscan 1172 which is an ex vivo/in vitro Micro-CT commercialized by the company Bruker.
